为了解胃癌组织中多种基因蛋白异常表达及意义,应用免疫组化技术检测51例手术切除胃癌组织标本中增殖细胞核抗原(PCNA)及P53、K-ras、Bcl-2蛋白的表达。结果胃癌组织中PCNA标记指数(PCNALI)为51.2±20.5,P53,K-ras,Bcl-2蛋白表达率分别37.3%,17.6%和68.6%,均显著高于正常胃粘膜(P<0.05)。PCNALI与肿瘤大小、淋巴结转移及临床分期密切相关(P<0.05)。肠型胃癌中K-ras、Bcl-2蛋白表达率分别为43.8%和93.8%,均显著高于胃型胃癌的6.0%(P<0.01),54.5%(P<0.05),而PCNALI及P53蛋白表达率两型胃癌无显著差别。提示PCNA、P53、K-ras、Bcl-2蛋白异常表达与胃癌的发生发展有关。PCNALI可能对判断胃癌的预后有一定价值。K-ras、Bcl-2异常表达可能与肠型胃癌发生有关
To understand the abnormal expression and significance of various gene proteins in gastric cancer, immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen (PCNA) and P53, K-ras and Bcl-2 protein in 51 specimens of surgically resected gastric cancer. Results The PCNA index was 51.2±20.5 in gastric cancer tissues, and the protein expression rates of P53, K-ras and Bcl-2 were 37.3%, 17.6% and 68.6%, respectively. In the normal gastric mucosa (P <0.05). PCNALI was closely related to tumor size, lymph node metastasis, and clinical stage (P<0.05). The expression rates of K-ras and Bcl-2 protein in intestinal type gastric cancer were 43.8% and 93.8%, respectively, which were significantly higher than those in gastric type gastric cancer (6.0%, P<0.01), 54.5%. (P<0.05), but there was no significant difference in the expression rates of PCNALI and P53 protein between the two types of gastric cancer. It is suggested that the abnormal expression of PCNA, P53, K-ras and Bcl-2 proteins is related to the occurrence and development of gastric cancer. PCNALI may have some value in determining the prognosis of gastric cancer. Aberrant expression of K-ras and Bcl-2 may be related to intestinal type gastric cancer
