目的:探讨广西莪术多糖(CKP)对2型糖尿病(T2DM)大鼠血糖的影响。方法:采用高糖高脂联合腹腔小剂量注射链脲佐菌素(STZ,45 mg·kg-1)的方法诱导T2DM模型,将造模成功的糖尿病大鼠随机分为4组:糖尿病模型组,CKP低、中、高剂量组(0.5,1.0,2.5 g·kg-1),另取10只正常大鼠设为正常组。给药组每天ig给药1次,连续给药6周;模型组和正常组则ig给予同体积的生理盐水。给药6周后,空腹检测大鼠血糖(FBG),胰岛素(INS),总胆固醇(TC),甘油三酯(TG)的水平;通过观察胰腺HE染色及胰腺免疫组化;检测胰岛β细胞Fas蛋白的表达。结果:与正常组大鼠相比,模型组大鼠血糖,TG,TC水平明显增高,且胰腺Fas蛋白表达增高,均具有统计学意义(P<0.05,P<0.01);与模型组比较,CKP给药组均可降低糖尿病大鼠的血糖,TC和TG水平;减少胰腺Fas蛋白表达,减少胰岛β细胞凋亡(P<0.05,P<0.01)。结论:广西莪术多糖降低2型糖尿病大鼠血糖,其作用可能是通过保护胰岛β细胞,减少其凋亡有关。
Objective: To investigate the effect of curcuma zedoariae polysaccharide (CKP) on blood glucose in type 2 diabetes mellitus (T2DM) rats. Methods: T2DM models were induced by intraperitoneal injection of streptozotocin (STZ, 45 mg · kg-1) in high glucose and high fat, and diabetic rats were randomly divided into 4 groups: diabetic model group , CKP low, middle and high dose group (0.5,1.0,2.5 g · kg-1), another 10 normal rats were set as normal group. Administration group administered ig 1 times a day, continuous administration for 6 weeks; model group and normal group ig given the same volume of saline. Six weeks after the administration, the levels of FBG, INS, TC and TG in rats were determined by fasting, HE staining and pancreas immunohistochemistry were performed to observe the pancreatic β-cell Fas protein expression. Results: Compared with the normal group, the levels of blood glucose, TG and TC in the model group were significantly increased, and the expression of Fas protein in the pancreas was significantly increased (P <0.05, P <0.01). Compared with the model group, The levels of TC, TG and TG in diabetic rats decreased significantly, while the expression of Fas protein in pancreatic tissues decreased and the apoptosis of pancreatic β cells decreased (P <0.05, P <0.01). CONCLUSION: Curcumin reduces the blood glucose in type 2 diabetic rats, which may be related to the protection of pancreatic islet β cells and their apoptosis.
