目的探讨益气养阴、祛瘀明目中药复方贞芪胶囊对糖尿病视网膜病变(Diabetic retinopathy,DR)大鼠血糖及视网膜中醛糖还原酶基因(Aldose Reductase,AR)mRNA表达的影响。方法高脂饮食喂养后采用脲链佐菌素(Streptozotocin,STZ)注射大鼠腹腔诱发糖尿病。在糖尿病大鼠病程3个月出现DR病理变化后用贞芪胶囊治疗,连续灌胃3个月,通过血糖的测定及逆转录-聚合酶链反应(RT-PCR)检测,探讨贞芪胶囊对DR大鼠血糖及视网膜中醛糖还原酶基因mRNA表达的影响。结果高脂饮食加链脲佐菌素诱导后3个月大鼠糖尿病视网膜病变造模成功;造模后及治疗前STZ大鼠血糖与空白对照组相比明显升高,差异有统计学意义(P<0.01);贞芪胶囊高、中、低剂量组AR mRNA水平均低于模型组和空白对照组,与模型组及空白对照组比较,差异均有统计学意义(P<0.01)。结论贞芪胶囊可降低DR大鼠视网膜中醛糖还原酶基因mRNA表达,同时有一定降低血糖的作用。
Objective To investigate the effects of Yiqiyangyin and Zhimingmudi compound compound Zhenqi capsules on blood glucose and aldose reductase (AR) mRNA expression in diabetic retinopathy (DR) rats. Methods Rats were injected with Streptozotocin (STZ) into the abdominal cavity to induce diabetes after high fat diet. After the pathological changes of DR in diabetic rats occurred 3 months after treatment, they were treated with Zhenqi capsules, and they were continuously fed for 3 months. The determination of blood glucose and reverse transcription-polymerase chain reaction (RT-PCR) were used to investigate the effects of Zhenqi capsules. Effects of aldose reductase gene mRNA expression in blood glucose and retina of DR rats . Results The model of diabetic retinopathy in rats was induced 3 months after induction with high-fat diet and streptozotocin. The blood glucose of STZ rats after modeling and before treatment was significantly higher than that of the blank control group, and the difference was statistically significant ( P<0.01); The AR mRNA levels in the high, middle and low doses of Zhenqi capsules were lower than those in the model group and the blank control group, and there were significant differences between the model group and the blank control group (P<0.01). Conclusion Zhenqi Capsule can reduce the expression of aldose reductase gene mRNA in the retina of DR rats, and it can also reduce the blood glucose.
