采用SRAP分子标记对70份万寿菊杂交后代优选株系基因组DNA进行遗传多样性分析,结果表明,9对SRAP引物共扩增得到250个位点和110个多态性位点,多态位点比例为44.0%;各引物组合Shannon多样性指数(Ⅰ)在0.054~0.466之间,平均为0.228;各引物组合的鉴别能力存在较大差异,以me4/em17、me6/em4最佳,可分别鉴定31个和42个株系。各株系获得的条带数介于168~217之间,平均为203.5条,其中多态性条带数介于22~71之间,平均为57.5条,占平均条带数的28.07%;Ⅰ指数介于0.655~0.693之间,平均为0.676。检测到株系间遗传距离变化范围为0.030~0.180,平均为0.098,变异系数介于17.81%~30.98%,平均为23.97%。亲缘关系聚类分析将所有株系在遗传相似性系数0.89处分为9类,其中株系41-3、45-1、36-2、6-1、26-2、16-1单独聚成一类,4-3、5-1(3)和35-2聚成一类,28-1、36-1和37-3聚成一类,其余58个株系聚成一类。
SRAP molecular markers were used to analyze the genetic diversity of the 70 preferred strains of marigold hybrids. The results showed that 9 pairs of SRAP primers co-amplified 250 loci and 110 polymorphic loci (44.0%). The Shannon index (Ⅰ) of each primer combination ranged from 0.054 to 0.466 with an average of 0.228. There was a significant difference in the discriminatory power of each primer combination, with me4 / em17 and me6 / em4 being the best 31 and 42 lines were identified. The number of bands obtained from each strain ranged from 168 to 217, with an average of 203.5. The number of polymorphic bands ranged from 22 to 71 with an average of 57.5, accounting for 28.07% of the average number of bands. Ⅰ index was between 0.655 ~ 0.693, with an average of 0.676. The detected genetic distance ranged from 0.030 to 0.180 with an average of 0.098. The coefficient of variation ranged from 17.81% to 30.98% with an average of 23.97%. The phylogenetic tree was used to classify all the strains into 9 groups at the genetic similarity coefficient of 0.89, in which the strains 41-3, 45-1, 36-2, 6-1, 26-2 and 16-1 were clustered together , 4-3, 5-1 (3) and 35-2, 28-1, 36-1 and 37-3 were clustered together, and the remaining 58 lines clustered into one.
