目的研究宫颈癌及相关组织中自噬基因Beclin-1、LC3-B的表达情况,并分析其与患者年龄、组织分化、临床期别、淋巴结转移状况等临床病理因素的关系。方法选取2014年3月-2016年3月西安交通大学第二附属医院124例石蜡包埋宫颈(癌)组织,包括80例宫颈鳞状细胞癌、24例宫颈腺癌、20例因子宫肌瘤或子宫腺肌病等原因切除的正常宫颈组织或炎性宫颈组织。制作宫颈(癌)组织芯片,量子点免疫荧光组织化学技术检测Beclin-1、LC3-B蛋白的表达,结合临床病理参数进行统计学分析。结果本研究观察到Beclin-1、LC3-B阳性信号在胞浆着色,相对于宫颈正常的上皮组织,Beclin-1、LC3-B阳性信号在宫颈癌中表达下降,差异有统计学意义(P<0.05)。宫颈鳞状细胞癌中,Beclin-1、LC3-B蛋白表达与宫颈癌患者年龄、组织学级别、肿瘤分期及淋巴结转移等临床病理参数均无明显相关性(均P>0.05)。35例Beclin-1阳性表达的宫颈鳞状细胞癌组织中,检测出20例LC3-B阳性表达;45例Beclin-1阴性表达的宫颈鳞状细胞癌组织中,检测出33例LC3-B阴性表达。提示Beclin-1蛋白表达与LC3-B蛋白表达呈正相关(r=0.309,P=0.005)。结论宫颈癌细胞中Beclin-1、LC3-B的表达下降与宫颈癌的发生发展有关。
Objective To study the expression of autophagy genes Beclin-1 and LC3-B in cervical cancer and related tissues and to analyze their relationship with clinical pathological factors such as age, histological differentiation, clinical stage and lymph node metastasis. METHODS: From March 2014 to March 2016, 124 patients with paraffin-embedded cervical (cancerous) tissues from the Second Affiliated Hospital of Xi’an Jiaotong University including 80 cases of cervical squamous cell carcinoma, 24 cases of cervical adenocarcinoma and 20 cases of uterine fibroids Or adenomyosis and other reasons to remove the normal cervical tissue or inflammatory cervical tissue. The cervical (cancer) tissue microarray was made. The expression of Beclin-1, LC3-B protein was detected by immunofluorescence histochemistry with quantum dots. Statistical analysis was made with clinicopathological parameters. Results The positive staining of Beclin-1 and LC3-B in the cytoplasm was observed in this study. The expression of Beclin-1 and LC3-B positive signals in cervical epithelium was significantly lower than that in normal cervical epithelium (P <0.05). Cervical squamous cell carcinoma, Beclin-1, LC3-B protein expression and cervical cancer patients age, histological grade, tumor stage and lymph node metastasis and other clinicopathological parameters were not significantly correlated (all P> 0.05). Among 35 cases of Beclin-1 positive cervical squamous cell carcinoma, 20 cases of LC3-B positive expression were detected. In 45 cases of Beclin-1 negative cervical squamous cell carcinoma, 33 cases of LC3-B negative expression. The results suggested that the expression of Beclin-1 protein was positively correlated with the expression of LC3-B protein (r = 0.309, P = 0.005). Conclusion The decreased expression of Beclin-1 and LC3-B in cervical cancer is related to the occurrence and development of cervical cancer.
