目的对主产区远志种质资源进行遗传多样性研究。方法利用ISSR分子标记分析来自于主产区陕西、山西和河北3个省的10个居群的远志种质资源的遗传多样性。结果 12条ISSR引物扩增出276条带,其中有271条为多态性条带,聚类分析结果表明,地理来源相同的野生居群和栽培居群先行聚合。野生和栽培远志居群的遗传变异主要发生在居群间,栽培居群的遗传多样性略低于野生居群,二者均不符合距离隔离模型。结论聚类分析结果与栽培居群多为就地取材的事实相符。造成远志这种遗传变异型式的原因在于其本身的繁殖生物学特性以及当前的粗放栽培模式。
Objective To study the genetic diversity of Polygalaceae germplasm resources in main producing areas. Methods ISSR molecular markers were used to analyze the genetic diversity of Polygala tenuifolia germplasm resources from ten populations in three main provinces Shaanxi, Shanxi and Hebei. Results A total of 276 bands were amplified by 12 ISSR primers, of which 271 were polymorphic bands. The results of cluster analysis indicated that wild populations and cultivated populations with the same geographical origin polymerized first. The genetic variation of wild and cultivated Polygala tenuiflora mainly occurred among populations, but the genetic diversity of cultivated populations was slightly lower than that of wild populations, both of which did not fit the distance isolation model. Conclusion The result of cluster analysis is consistent with the fact that cultivated population is mostly local. The reason for this genetic variant of Polygalaceae lies in its own reproductive biology and current extensive cultivation patterns.
