目的 分析比较5种WRI1转基因稻米粗脂肪表达效果.方法 将椰子中的调节脂肪酸合成基因-WRI1基因导入粳稻品种中花11中,获得再生植株ZZWR1、ZZWR2、ZZWR3、ZZWR4和ZZWR5,用索氏提取法提取转基因稻米及其对照稻米中的脂肪,再将粗脂肪进行脂肪酸甲酯化,并用气相色谱法-质谱联用法(GC-MS)对其进行定性定量测定.结果 与对照组的粗脂肪含量2.59％相比,ZZWR1、ZZWR2、ZZWR4的粗脂肪含量范围为2.62％ ～2.77％,其中ZZWR4的粗脂肪含量最高(2.77％),RSD=0.96％,与对照组相比差异有统计学意义(P＜0.01).GC-MS测定棕榈酸甲酯、硬脂酸甲酯和油酸甲酯含量,其中油酸甲酯的含量最高.结论 椰子中的WRI1基因的导入可提高稻米脂肪含量,但不同WRI1基因的插入位点可能会影响原有脂肪酸的表达,ZZWR4植株插入基因位点比较得当,具有良好的基因表达效果.
