目的研究人间布病患者外周血单个核细胞中TLR2和TLR4的mRNA表达量和血清中IL-2、IL-6、TNF-α的表达水平。方法采用qRT-PCR技术检测布病患者和健康对照组外周血单个核细胞TLR2和TLR4的mRNA表达量;同时采用ELISA方法检测血清中IL-2、IL-6、TNF-α的浓度。结果急性期布病患者组TLR2、TLR4的mRNA表达量高于慢性期及健康对照组(P均<0.01);慢性期TLR2、TLR4的mRNA表达量与健康对照组比较无统计学差异(P均>0.05);布病急、慢性期患者组血清中IL-2的浓度均低于健康对照(P均<0.05),急性期患者血清中IL-2的浓度高于慢性期(P<0.05);急、慢性期布病患者血清中IL-6和TNF-α的浓度均高于健康对照(P均<0.05),且急性期患者血清中IL-6和TNF-α的浓度高于慢性期(P均<0.05)。结论 TLR2、TLR4及IL-2、IL-6和TNF-α可能参与了机体抗布鲁氏菌病的免疫反应,在布鲁氏菌病的发生发展中起重要作用,其动态变化能反映布鲁氏菌病的病情发展及转归。
Objective To study the mRNA expression levels of TLR2 and TLR4 and the levels of IL-2, IL-6 and TNF-α in peripheral blood mononuclear cells of human brucellosis patients. Methods qRT-PCR was used to detect the mRNA expression of TLR2 and TLR4 in peripheral blood mononuclear cells of patients with brucellosis and healthy controls. Meanwhile, the concentrations of IL-2, IL-6 and TNF-α in serum were detected by ELISA. Results The mRNA expressions of TLR2 and TLR4 in patients with acute periodontal disease were significantly higher than those in chronic phase and healthy controls (all P <0.01). There was no significant difference in the expression of TLR2 and TLR4 mRNA between the two groups > 0.05). Serum levels of IL-2 in patients with acute and chronic IBD were lower than those in healthy controls (all P <0.05), IL-2 levels in serum were higher than those in chronic phase (P <0.05) . The concentrations of IL-6 and TNF-α in serum of patients with acute and chronic bursal disease were significantly higher than those of healthy controls (all P <0.05), and the levels of IL-6 and TNF-α in serum of patients with acute and chronic phase were higher than those in chronic phase (P <0.05). Conclusion TLR2, TLR4, IL-2, IL-6 and TNF-α may play an important role in the pathogenesis of Brucellosis and their dynamic changes may reflect the immune response of the body to brucellosis The development of the disease and prognosis of brucellosis.
