目的 研究神经细胞粘附分子(NCAM)与基质金属蛋白酶(MMP)在人脑星形细胞瘤中的作用。方法 用原位杂交和免疫组织化学方法分别检测40例人脑星形细胞瘤组织中的NCAMmRNA及MMP-2、9蛋白的表达。结果 星形细胞瘤Ⅰ、Ⅱ级中NCAM-140mRNA,MMP-2、9蛋白的表达阳性数为16/17,12/17,5/17,病理指数为 29.08±12.31,19.63±11.37,13.32±11.53,Ⅲ、Ⅳ级中表达阳性数为11/23,22/23,17/23,病理指数为15.02±10.41,34.52±7.65,30.29±16.88,人脑星形细胞瘤高级别(Ⅲ、Ⅳ级)与低级别(Ⅰ、Ⅱ级)差异显著。MMP-2蛋白与NCAMmRNA两者有显著负相关性。结论NCAMmRNA及MMP-2、9蛋白表达与星形细胞瘤的恶性程度有关。NCAM和MMP对星形细胞瘤的发生发展与侵袭转移起重要作用。
Objective To investigate the role of neural cell adhesion molecule (NCAM) and matrix metalloproteinase (MMP) in human astrocytoma. Methods In situ hybridization and immunohistochemistry were used to detect the expression of NCAMmRNA and MMP-2, 9 in 40 human astrocytoma tissues. Results The positive expression rates of NCAM-140 mRNA and MMP-2, 9 in grade Ⅰ and grade Ⅱ were 16/17, 12/17 and 5/17 respectively, and the pathological indexes were 29.08 ± 12.31, 19.63 ± 11.37 and 13.32 ± The positive numbers in the group of 11.53, Ⅲ and Ⅳ were 11 / 23,22 / 23,17 / 23 and the pathological indexes were 15.02 ± 10.41,34.52 ± 7.65,30.29 ± 16.88. The high grade of human astrocytoma (Ⅲ, Ⅳ Grade) and low level (Ⅰ, Ⅱ grade) significant difference. There was a significant negative correlation between MMP-2 protein and NCAM mRNA. Conclusions The expression of NCAM mRNA and MMP-2, 9 protein is related to the malignancy of astrocytomas. NCAM and MMP play an important role in the development, invasion and metastasis of astrocytoma.
