目的建立大鼠血浆中京尼平苷酸、原儿茶酸、绿原酸、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷的UPLC-MS/MS测定方法,并进行五种成分在大鼠体内药代动力学研究。方法大鼠静脉注射杜仲提取物后,采用甲醇沉淀血浆蛋白,以葛根素为内标,采用Waters BEH C18(2.1 mm×100 mm,1.7μm)色谱柱,0.1%甲酸乙腈-0.1%甲酸水溶液梯度洗脱,采用电喷雾电离源(ESI),以多反应监测(MRM)方式进行检测。结果京尼平苷酸等五种成分在血浆中呈良好线性关系。方法学考察均符合要求,日内、日间精密度、准确度和稳定性良好,提取回收率86.06%~104.39%,无基质效应。京尼平苷酸等5个指标成分在大鼠体内消除较快。结论建立的HPLC-MS/MS测定方法能快速灵敏地检测生物样品中京尼平苷酸、原儿茶酸、绿原酸、松脂醇二葡萄糖苷和松脂醇单葡萄糖苷的血药浓度,可用于五种成分的药代动力学研究。
OBJECTIVE To establish a UPLC-MS / MS method for the determination of geniposidic acid, protocatechuic acid, chlorogenic acid, pinoresinol diglucoside, and pinoresinol mono-glucoside in rat plasma and five components in rat in vivo Study of generation dynamics.
