目的研究银耳提取物对大鼠抗抑郁活性的影响。方法采用雄性Wistar大鼠(体重180~220 g)建立慢性应激抑郁症大鼠模型,建模成功大鼠随机分为4组,分别为模型组(生理盐水2.0 ml/kg),银耳提取物低剂量组(0.5 g/kg),银耳提取物中剂量组(1.0 g/kg),银耳提取物高剂量组(1.5 g/kg),取空白未建模大鼠作为空白组,给予等体积的生理盐水,五组大鼠连续灌胃给药28 d,观察大鼠悬尾不动时间和游泳不动时间,并测定大鼠血浆及下丘脑神经递质多巴胺(DA)、去甲肾上腺素(NE)含量。结果 1.5 g/kg银耳提取物可以显著提高抑郁大鼠体重(P<0.01),增加大鼠蔗糖偏爱度(P<0.01),同时1.0~1.5 g/kg银耳提取物剂量依赖性的减少大鼠悬尾不动时间和游泳不动时间(P<0.05),增加大鼠血浆及下丘脑多巴胺(DA)含量(P<0.05)。1.5 g/kg银耳提取物显著提高大鼠血浆及下丘脑去甲肾上腺素(NE)含量(P<0.01)。结论银耳提取物具有显著的抗抑郁活性,其作用可能与其增加大鼠血浆及下丘脑神经递质含量相关。
Objective To study the effects of Tremella fuciformis extract on the antidepressant activity in rats. Methods A rat model of chronic stress depression was established in male Wistar rats (body weight 180-220 g). The rats were randomly divided into 4 groups: model group (saline 2.0 ml / kg), Tremella fuciformis Low dose group (0.5 g / kg), white fungus extract medium dose group (1.0 g / kg), Tremella fuciformis extract high dose group (1.5 g / kg), take blank unmodelled rats as blank group, . The rats in the five groups were given gavage for 28 days continuously to observe the immobility time and swimming immobility time of the rats and the changes of plasma and neurotransmitter dopamine (DA), norepinephrine (NE) content. Results Tremella extract (1.5 g / kg) significantly increased body weight (P <0.01) and increased sucrose preference (P <0.01) in rats, while a dose-dependent decrease in rats treated with 1.0-1.5 g / kg tremella Hang tail time and swimming time (P <0.05), and increase the content of dopamine (DA) in the plasma and hypothalamus of rats (P <0.05). The extract of Tremella fuciformis with 1.5 g / kg significantly increased the content of norepinephrine (NE) in the plasma and hypothalamus (P <0.01). Conclusion Tremella fuciformis has significant antidepressant activity, which may be related to the increase of neurotransmitters in plasma and hypothalamus of rats.
