目的研究原花青素对肿瘤坏死因子-α(TNF-α)诱导HeLa细胞凋亡和caspase-8活化的影响。方法采用流式细胞术检测细胞凋亡,荧光检测试剂盒测定caspase-8活性,Westernblot检测caspase-8酶原表达水平。结果原花青素(5、10、20mg/L)对重组人肿瘤坏死因子-α(rhTNF-α,100μg/L)诱导的HeLa细胞凋亡均有显著的抑制作用(P<0.01),呈剂量依赖关系;原花青素(5、10、20mg/L)对rhTNF-α(100μg/L)诱导的HeLa细胞caspase-8活化均有显著抑制作用(P<0.01),呈剂量依赖关系;原花青素(5、10、20mg/L)对HeLa细胞caspase-8酶原表达无明显影响。结论原花青素抑制TNF-α诱导HeLa细胞凋亡可能与其抑制caspase-8活化有关。
Objective To investigate the effect of procyanidins on the apoptosis and caspase-8 activation of HeLa cells induced by tumor necrosis factor-α (TNF-α). Methods Cell apoptosis was detected by flow cytometry, caspase-8 activity was measured by fluorescence detection kit, and caspase-8 zymogen expression was detected by Western blot. Results Proanthocyanidins (5, 10, 20 mg/L) significantly inhibited the apoptosis of HeLa cells induced by recombinant human tumor necrosis factor-α (rhTNF-α, 100 μg/L) (P<0.01) in a dose-dependent manner. Proanthocyanidins (5, 10, 20 mg/L) significantly inhibited the activation of caspase-8 in HeLa cells induced by rhTNF-α (100 μg/L) (P<0.01) in a dose-dependent manner; procyanidins (5,10, 20mg/L) had no significant effect on the expression of caspase-8 proenzyme in HeLa cells. Conclusion The inhibition of TNF-α-induced apoptosis of HeLa cells by procyanidins may be related to its inhibition of caspase-8 activation.
