利用重组RTPCR方法从人胎盘组织中扩增多重肿瘤抑制基因(MTS1)全长cDNA片断,克隆测序后,亚克隆入哺乳动物高效表达质粒pCEP中。将表达质粒转染两种遗传背景不同的宫颈癌细胞系,发现外源基因MTS1/p16基因的导入对HPV阳性的宫颈癌细胞系具有明显生长抑制作用,并出现细胞滞留在G1期的特性
Recombinant RTPCR method was used to amplify the full-length cDNA of MTS1 from human placenta tissue, cloned and sequenced, and then subcloned into mammalian high expression plasmid pCEP. The expression plasmids were transfected into two cervical cancer cell lines with different genetic backgrounds. It was found that the introduction of the exogenous gene MTS1/p16 gene has a significant growth inhibitory effect on HPV-positive cervical cancer cell lines, and the characteristics of cell retention in G1 phase.
