沉默白细胞相关免疫球蛋白样受体1(LAIR-1)在卵巢癌H08910细胞的表达,探讨其对卵巢癌细胞增殖功能的影响。构建LAIR-1的shRNA慢病毒表达载体,转染人浆液性卵巢癌细胞系H08910。RT-PCR和Western blot检测H08910细胞中LAIR-1分子的沉默效率;采用PI染色法和MTT法检测沉默LAIR-1表达对HO8910细胞周期及增殖功能的影响。成功建立沉默LAIR-1分子表达的稳定HO8910细胞株。与阴性对照组比较,转染LAIR-1-shRNA慢病毒载体的HO8910细胞中LAIR-1的mRNA及蛋白水平表达均明显降低(P<0.05)。LAIR-1-shRNA组G1期细胞数目明显低于CON组和NCshRNA组(P<0.05);沉默LAIR-1分子表达后,LAIR-1-shRNA HO8910细胞的增殖率明显高于NC-shRNA HO8910组和空白对照组(P<0.05)。沉默卵巢癌细胞HO8910LAIR-1分子表达可明显增强细胞增殖的能力,提示卵巢癌细胞表达的LAIR-1可能对癌细胞的生物学功能发挥抑制作用。
To investigate the effect of silencing leukocyte associated immunoglobulin-like receptor 1 (LAIR-1) in ovarian cancer cell line H08910 and its effect on the proliferation of ovarian cancer cells. LAIR-1 shRNA lentivirus expression vector was constructed and transfected into human serous ovarian cancer cell line H08910. The silencing efficiency of LAIR-1 in H08910 cells was detected by RT-PCR and Western blot. The effect of silenced LAIR-1 expression on cell cycle and proliferation of HO8910 cells was detected by PI staining and MTT assay. A stable HO8910 cell line expressing silenced LAIR-1 molecule was successfully established. Compared with the negative control group, the expression of LAIR-1 mRNA and protein in HO8910 cells transfected with LAIR-1-shRNA lentiviral vector significantly decreased (P <0.05). The number of G1 phase cells in LAIR-1-shRNA group was significantly lower than that in CON group and NCshRNA group (P <0.05). The proliferation rate of LAIR-1-shRNA HO8910 cells was significantly higher than that of NC-shRNA HO8910 cells And blank control group (P <0.05). The expression of HO8910LAIR-1 in the silenced ovarian cancer cells can significantly enhance the cell proliferation, suggesting that LAIR-1 expressed in ovarian cancer cells may have an inhibitory effect on the biological function of cancer cells.
