目的 :探讨良恶性骨肿瘤细胞 p2 7蛋白表达与 DNA含量的关系及意义。方法 :取骨肿瘤新鲜标本 5 2例(骨软骨瘤 10例 ,骨巨细胞瘤 10例 ,骨肉瘤 2 0例 ,其他恶性骨肿瘤 12例 ) ,运用流式细胞术、单克隆抗体技术和定量免疫荧光技术 ,检测瘤细胞 p2 7蛋白表达、S期细胞百分数 (SPF)及 DNA含量 (DI) ,分析三者之间的关系。结果 :5 2例骨肿瘤 p2 7蛋白表达的平均 FI值为 1.37(骨软骨瘤 2 .2 4± 0 . 38,骨巨细胞瘤 1.79± 0 .36 ,骨肉瘤1.0 6± 0 .39,其他恶性骨肿瘤 1.0 1± 0 .45 ) ;p2 7<1.37组 2 5例 ,均为恶性骨肿瘤 ;p2 7<1.37组的 DI、SPF均明显高于 p2 7≥ 1.37组 (P<0 .0 0 1)。结论 :p2 7蛋白表达减少 ,导致细胞周期失调 ,S期细胞增多 ,DNA合成增强 ,细胞过度增生、恶变 ,可能是骨肿瘤发生发展的原因之一。
Objective: To investigate the relationship between p27 protein expression and DNA content in benign and malignant bone tumor cells. Methods: Fifty-two fresh specimens of bone tumors were taken (10 osteochondromas, 10 with giant cell tumor of bone, 20 with osteosarcoma, and 12 with other malignant bone tumors). Flow cytometry, monoclonal antibody technology and quantification were used. Immunofluorescence technique was used to detect the expression of p27 protein in tumor cells, percentage of S phase cells (SPF) and DNA content (DI), and analyze the relationship among the three. Results: The mean FI value of p2-7 protein expression in 52 cases of bone tumors was 1.37 (2.24±0.38 for osteochondroma, 1.79±0.36 for giant cell tumor of bone, 1.06±0.39 for osteosarcoma, others. Malignant bone tumors 1.0 1 ± 0 .45 ); p2 7 <1.37 group of 25 cases, all malignant bone tumors; p2 7 <1.37 group DI, SPF were significantly higher than p2 7 ≥ 1.37 group (P <0. 0 0 1). Conclusion : The decrease of p27 protein expression leads to the disorder of cell cycle, the increase of S phase cells, the increase of DNA synthesis, and the excessive proliferation and malignant transformation of cells, which may be one of the causes of bone tumor development.
