目的探讨碱性成纤维细胞生长因子(bFGF)促进骨髓间充质干细胞(BMSCs)细胞增殖与分化的作用及其可能机制。方法从SD大鼠骨髓中获得BMSCs,纯化培养传代后用不同浓度的bFGF诱导,经MTT法检测bFGF对BMSCs生长的影响,应用透射电镜和光镜观察细胞形态结构变化,免疫细胞化学染色显示GFAP表达,Western blot方法分析p-ERK蛋白表达。结果 bFGF对BMSCs具有促增殖作用,在倒置光显微镜和透射电镜下可见到有神经胶质样细胞形成,免疫细胞化学证明GFAP的表达较对照组显著增高(P<0.01),Western blot证明bFGF上调ERK蛋白表达(P<0.05)。结论 bFGF可活化Ras/ERK信号转导通路,促进BMSCs细胞增殖分化为GFAP阳性的神经胶质样细胞。
Objective To investigate the effect of basic fibroblast growth factor (bFGF) on the proliferation and differentiation of bone marrow mesenchymal stem cells (BMSCs) and its possible mechanism. Methods BMSCs were obtained from bone marrow of SD rats. After passage of culture, the cells were induced with different concentrations of bFGF. MTT assay was used to detect the effect of bFGF on the growth of BMSCs. Transmission electron microscopy and light microscopy were used to observe the changes of cell morphology. Immunocytochemical staining showed GFAP expression Western blot analysis of p-ERK protein expression. Results bFGF had a proliferative effect on BMSCs. Glial-like cells were found under inverted light microscope and transmission electron microscope. Immunocytochemistry showed that GFAP expression was significantly higher than that of the control group (P <0.01). Western blot showed that bFGF was up-regulated ERK protein expression (P <0.05). Conclusion bFGF can activate Ras / ERK signal transduction pathway and promote the proliferation and differentiation of BMSCs into GFAP positive glial like cells.
