目的 :探讨急性缺血诱导大鼠心肌细胞凋亡的时间演变和空间分布规律。方法 :以DNA电泳和TUNEL分析检测SD大鼠心肌的细胞凋亡情况。结果 :缺血 2h开始出现DNA梯形条带和TUNEL阳性细胞 ,DNA条带和凋亡指数随缺血时间延长而显著增加 (P <0 0 1)。TUNEL阳性心肌细胞在缺血区散在分布 ,缺血边缘区凋亡指数显著高于缺血中央区 (P <0 0 5 ) ,缺血区内膜下凋亡指数显著高于外膜下 (P <0 0 1)。结论 :在本实验设定的时间范围内 ,急性缺血诱导的心肌细胞凋亡与缺血时间具有时效关系 ;调亡的心肌细胞在缺血区散在分布 ,以缺血区两侧边缘和内膜下心肌多见。
Objective: To investigate the time evolution and spatial distribution of acute cardiomyocyte apoptosis induced by acute ischemia in rats. Methods: The apoptosis of SD rat myocardium was detected by DNA electrophoresis and TUNEL assay. Results: The DNA ladder and TUNEL positive cells appeared at 2h after ischemia, and the DNA bands and apoptosis index increased significantly with prolongation of ischemia time (P <0.01). TUNEL-positive cardiomyocytes were scattered in the ischemic area, the apoptotic index in ischemic marginal zone was significantly higher than that in the central ischemic area (P <0.05), and the apoptosis index in ischemic subintimal area was significantly higher than that in the subepithelial (P <0 0 1). CONCLUSION: Apoptosis of cardiomyocytes induced by acute ischemia has a time-dependent relationship with ischemic time within the time frame set in this experiment. The apoptotic cardiomyocytes are scattered in the ischemic area, Myocardium more common.
