目的探讨环氧酶-2蛋白与细胞DNA损伤的关系。方法采用大鼠大脑中动脉阻塞脑缺血模型,应用HE染色观察缺血损伤的演变,免疫组化染色观察环氧酶-2蛋白的表达,DNA末端标记法(TUNEL)法观察细胞DNA损伤。结果环氧酶-2蛋白的表达与TUNEL阳性细胞在时程上一致,解剖部位上重叠。结论环氧酶-2是诱导细胞凋亡的重要因素,脑缺血后环氧酶-2的高度表达促进了便死范围的扩大。
Objective To investigate the relationship between cyclooxygenase-2 protein and cell DNA damage. Methods The model of occlusion of middle cerebral artery occlusion in rats was made. The development of ischemic injury was observed by HE staining. The expression of cyclooxygenase - 2 protein was observed by immunohistochemical staining. The DNA damage was observed by TUNEL method. Results The expression of cyclooxygenase-2 protein was consistent with the time course of TUNEL-positive cells and the anatomic sites overlapped. Conclusions Cyclooxygenase-2 is an important factor in inducing cell apoptosis. The high expression of cyclooxygenase-2 promotes the enlargement of the area of hemorrhage after cerebral ischemia.
