目的:检测胃动蛋白基因1(gastrokine 1,GKN1)在胃间质瘤和瘤旁组织中的表达,分析其与GIST生物学行为的关系,方法:选取30例川北医学院附属医院病理科胃GIST及瘤旁组织石蜡标本,应用免疫组化检测GKN1在胃GIST和瘤旁组织中的表达;收集20例川北医学院附属医院普外科手术并经病理及免疫组化确诊的胃GIST肿瘤组织及瘤旁新鲜组织标本,应用实时荧光定量聚合酶链反应(real time fluorescence quantitative polymerase chain reaction,RT-PCR)和Western blot的方法检测GKN1在GIST肿瘤组织和瘤旁组织中的表达情况,同时分析实验数据与临床资料的关系。结果:GKN1主要表达在胃粘膜组织,GKN1的表达在胃GIST中显著低于瘤旁组织(P<0.05),GKN1蛋白高度表达于极低危险组的胃GIST,GKN1 mRNA的表达与性别、年龄及核分裂像无差异(P>0.05),但胃GIST的GKN1 mRNA表达量:直径(≤5cm)组明显高于直径(>5cm)组(P<0.05),极/低危组显著高于中/高危组(P<0.05)。结论:GKN1可能在胃GIST中从极低危到高危的发展过程中起着一定的作用,对鉴别其生物学行为有一定的指示和参考作用。
OBJECTIVE: To detect the expression of gastrokine 1 (GKN1) in gastric stromal tumors and adjacent tissues and to analyze its relationship with the biological behavior of GIST.Methods: Thirty patients with pathological stomach of Affiliated Hospital of North Sichuan Medical College GIST and para-tumor paraffin specimens were collected. Immunohistochemistry was used to detect the expression of GKN1 in gastric GIST and paraneoplastic tissues. Twenty cases of gastric GIST tumor tissues were collected and confirmed by pathology and immunohistochemistry in 20 cases of North Sichuan Medical College Affiliated Hospital. The tumor tissue adjacent to fresh tissue was detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and Western blot method to detect the expression of GKN1 in GIST tumor tissues and adjacent tissues, while analysis of experimental The relationship between data and clinical data. Results: The expression of GKN1 was mainly in gastric mucosa. The expression of GKN1 in gastric GIST was significantly lower than that in adjacent tissues (P <0.05). The expression of GKN1 was highly expressed in gastric GIST in very low risk group. The expression of GKN1 mRNA was not related to gender, age (P> 0.05). However, the expression of GKN1 mRNA in gastric GIST was significantly higher than that in diameter (> 5cm) group (P <0.05) / High risk group (P <0.05). CONCLUSION: GKN1 may play a role in the development of gastric GIST from very low risk to high risk, and it may be an indicator and reference for the identification of its biological behavior.
