目的了解糖尿病模型大鼠心肌组织中心肌营养素-1(CT-1)蛋白的表达,探讨黄芪注射液改善糖尿病大鼠心肌病变的作用机制。方法雄性Wistar大鼠随机分为正常组8只及糖尿病造模组22只,糖尿病造模组成模后再随机分为模型组和黄芪组,每组11只。黄芪组予黄芪注射液8 g/(kg.d)灌胃8周。测定大鼠心脏指数和心肌纤维含量的变化,并用免疫组化法检测CT-1的表达。结果模型组和黄芪组血糖高于对照组(P<0.01);心脏指数(心脏重量/体重)模型组高于正常组(P<0.01),黄芪组心脏指数较模型组降低(P<0.05);Masson染色中以目标面积计算的胶原含量,模型组高于正常组(P<0.01),黄芪组比模型组减少(P<0.05);CT-1蛋白的表达模型组高于正常组(P<0.01),黄芪组较模型组减少(P<0.05)。结论 CT-1的过度表达参与了糖尿病心肌病变中心肌肥大的发病过程,黄芪注射液对CT-1的表达和心肌肥大有抑制作用。
Objective To investigate the expression of cardiac troponin-1 (CT-1) protein in myocardial tissue of diabetic rats and to explore the possible mechanism of Astragalus injection in improving cardiomyopathy in diabetic rats. Methods Male Wistar rats were randomly divided into normal group (n = 8) and diabetic model group (n = 22). Diabetic model rats were randomly divided into model group and Astragalus group (n = 11). Astragalus group Astragalus injection 8 g / (kg.d) gavage for 8 weeks. The changes of rat cardiac index and myocardial fiber content were determined. The expression of CT-1 was detected by immunohistochemistry. Results The blood glucose of the model group and the astragalus group was higher than that of the control group (P <0.01); the cardiac index (heart weight / body weight) model group was higher than the normal group (P <0.01) (P <0.05). The expression of CT-1 protein in the model group was higher than that in the normal group (P <0.01), and that in the Astragalus group was lower than that in the model group <0.01), Astragalus group than the model group decreased (P <0.05). Conclusion The overexpression of CT-1 is involved in the pathogenesis of myocardial hypertrophy in diabetic cardiomyopathy. Astragalus injection can inhibit the expression of CT-1 and myocardial hypertrophy.
