目的:研究氧化锌(ZnO)纳米颗粒对雌性小鼠的在体毒性作用,着重观察其对雌性小鼠生殖系统的影响。方法:成年雌性小鼠隔日腹腔注射ZnO纳米(200、500mg/kg),对照组注射等体积生理盐水,共给药5次。停药1周后,测量各重要脏器的脏器系数,血清生化指标、雌二醇和孕激素的水平,主要脏器作病理切片和HE染色。结果:与对照组相比,给药ZnO纳米(200mg/kg、500mg/kg)的小鼠,肝脏、肾脏和卵巢的脏器系数均显著增加,200mg/kg组的心脏脏器系数显著增加(P均<0.05);给药200mg/kg和500mg/kg组小鼠血生化指标中,尿酸(UA)显示增高,且有统计学意义(P<0.05),提示在200~500mg/kgZnO纳米作用下肾功能有异常;此外,给药200mg/kg和500mg/kg组小鼠的乳酸脱氢酶(LDH)增高明显,提示心肌也有轻度的损伤。给药200mg小鼠的雌二醇、孕酮水平没有统计学差异(P>0.05),生殖系统脏器组织病理也未发现明显异常。结论:200~500mg/kgZnO纳米对雌性小鼠肝、肾和心功能有轻度影响,对小鼠的雌孕激素的分泌及卵巢组织无明显影响。因此,200~500mg/kgZnO纳米对小剂量ZnO纳米有一定的毒性,而对雌性小鼠生殖系统功能是安全的。
OBJECTIVE: To study the in vivo toxicity of zinc oxide (ZnO) nanoparticles on female mice and its effect on the reproductive system of female mice. METHODS: Adult female mice were injected intraperitoneally with ZnO nanoparticles (200, 500mg / kg) every other day. The control group was injected with equal volume of normal saline for 5 times. One week after discontinuation, organ coefficient, serum biochemical index, estradiol and progesterone levels of major organs were measured. The pathological sections of major organs and HE staining were performed. Results: Compared with the control group, the organ coefficient of liver, kidney and ovary increased significantly in mice treated with ZnO nanoparticles (200mg / kg, 500mg / kg), and the coefficient of cardiac organ significantly increased in 200mg / kg group P <0.05). UA in serum of 200mg / kg and 500mg / kg group was significantly higher than that of control group (P <0.05), suggesting that 200 ~ 500mg / Lower kidney function abnormalities; In addition, administration of 200mg / kg and 500mg / kg group of mice lactate dehydrogenase (LDH) increased significantly, suggesting that there is mild myocardial damage. Administration of 200mg mouse estradiol, progesterone levels were not statistically different (P> 0.05), reproductive system, histopathology also found no significant abnormalities. Conclusion: 200 ~ 500mg / kg ZnO nanoparticles have a slight effect on liver, kidney and heart function in female mice, but have no significant effect on the secretion of estrogen and progesterone and ovarian tissue in mice. Therefore, 200 ~ 500mg / kg ZnO nanomaterials have a certain toxicity to small doses of ZnO nanomaterials, while the female mice reproductive system function is safe.
