本文报道了用β-丙内酯和紫外线(uv)照射灭活血浆、冷沉淀物和因子Ⅷ浓制剂中的HIV。用H9细胞培养HIV,通过测定第14-27天上清液中的逆转录酶活性或用ELISA测定HIV抗原和/或用免疫荧光试验测定细胞中的HIV抗原来检测病毒的复制情况。血浆先用25%(v/v)的新鲜β-丙内酯在pH7.2、23℃处理1小时,然后用256nm uv照射。冷沉淀物(3.5%)用0.05%β-丙内酯在pH7.2、23℃处理90分钟,纯化的因子Ⅷ浓制剂用uv进行照射。
Herein, it is reported that irradiation of inactivated plasma, cryoprecipitate, and HIV VIII concentrates with β-propiolactone and ultraviolet (uv) radiation. HIV cells were cultured with H9 cells and virus replication was determined by measuring reverse transcriptase activity in supernatants from day 14-27 or by assaying HIV antigens by ELISA and/or assaying HIV antigens in cells by immunofluorescence assays. Plasma was first treated with 25% (v/v) of fresh β-propiolactone at pH 7.2, 23°C for 1 hour and then irradiated with 256 nm uv. The cryoprecipitate (3.5%) was treated with 0.05% [beta]-propiolactone at pH 7.2, 23[deg.]C for 90 minutes, and the purified Factor VIII concentrate was irradiated with uv.
