目的 探讨 Rh血型 D抗原弱表现型 D抗原数目和 (或 ) D抗原表位数减少形成的分子基础。方法 常规 PCR技术检测 3名 D抗原弱表现型非血缘关系个体 D基因的第 4内含子 ,多重 PCR技术分析第 3、第 6和第 9外显子 ,并分别对 3名个体的 D基因部分序列进行分析。结果 3名 D抗原弱表现型个体 D基因第 4内含子 ,第 3、第 6和第 9外显子检测结果与正常 D基因完全一致 ,部分序列分析未发现基因突变和 D/CE基因交换。结论 D抗原数目和 (或 ) D抗原表位数的减少造成其血清学弱表现型可能与 D基因杂合性以外的其他因素有关
Objective To investigate the molecular basis of the formation of weakly expressed D antigen and / or reduced epitope of D antigen in Rh blood group D antigen. Methods The 4th intron of D gene of 3 individuals with weak phenotype of D-antigen was detected by conventional PCR. The 3rd, 6th and 9th exons were analyzed by multiplex PCR and the D gene of 3 individuals Part of the sequence analysis. Results The results of intron 4, 3, 6 and 9 exons in D gene of D-poor weak-expressive individuals were completely consistent with those of normal D gene. Partial sequence analysis showed no gene mutation and D / CE gene exchange . Conclusion The decrease of the number of D antigen and / or the number of D antigen epitopes may cause the low serum serotypes to be related to other factors besides heterozygosity of D gene
