目的:探索一种能应用于临床的、快速、准确地判定骨骼肌变性程度的方法。方法:以室温下保存的离断后人前臂屈侧骨骼肌为研究对象,采用荧光发光法测定肢体离断后每间隔1 h 骨骼肌中ATP的含量。同时观察骨骼肌在光镜、电镜下的亚显微结构变化。结果:人离断肢体骨骼肌中ATP含量与骨骼肌变性程度有明显的对应关系。当骨骼肌中ATP含量下降95% 以后,其电镜下发生了不可逆性损伤。结论:根据人骨骼肌中ATP含量的变化可以确切地判断肌肉的变性程度。而荧光发光方法快速、准确、稳定,可应用于临床,为判断离断肢体能否再植提供一种可信、可行的检测方法。
Objective: To explore a method that can be used clinically to determine the degree of skeletal muscle degeneration rapidly and accurately. Methods: The flexor skeletal muscle of the forearm of the severed forearm preserved at room temperature was used as the research object. The content of ATP in the skeletal muscle was measured by fluorescence spectrophotometry at 1 h intervals after the limbs were severed. At the same time, observe the changes of the sub-microstructure of skeletal muscle under light microscope and electron microscope. Results: There was a clear correspondence between ATP content in skeletal muscle and skeletal muscle degeneration in human limbs. When ATP content in skeletal muscle decreased by 95%, irreversible damage occurred under electron microscope. Conclusion: The degree of muscle degeneration can be accurately determined based on the change of ATP content in human skeletal muscle. The fluorescence luminescence method is rapid, accurate and stable and can be applied to clinic to provide a credible and feasible detection method for determining whether replantable limbs can be replanted.
