生物固氮能提高土壤肥力,改善作物氮素营养,一直是人们普通关心的问题.如何发挖更多的固氮微生物资源.增强其固氮作用,则是目前生物固氮研究中的一个方面.本文结合土壤资源普查,分离鉴定黑垆土中某些非共生固氮细菌,并测定其纯培养的固氮特性.一、材料和方法(一)样品采集我们从宁夏固原地区的城阳公社、古城公社和彭阳公社三个地点的黑垆土,分层采样,带回室内分析.(二)菌种分离为了能分离到较多的固氮细菌种类,我们采用了两个培养基,即尼古拉期无氮培养基和Line及Loutit介绍的无氮培养基.后一培养基有硫代乙醇酸钠,能使培养基本身造成较强的还原条件,因此固氮活性测定也采用了这一培养基.
Biological nitrogen fixation can improve soil fertility and improve crop nitrogen nutrition has always been a common concern.How to dig more nitrogen fixation microbial resources to enhance its role in nitrogen fixation is one of the aspects of the current biological nitrogen fixation.In this paper, Resources census, isolation and identification of some non-symbiotic nitrogen fixing bacteria in black soil, and to determine its pure culture of nitrogen fixation characteristics.Materials and methods (a) sample collection We from the Ningxia Guyang areaChengyang commune, the ancient city commune and Penang Commune three sites of black soil, stratified sampling, back to indoor analysis. (B) separation of bacteria in order to be able to separate more nitrogen-fixing bacteria species, we use two media, namely, Nicolais-free nitrogen Medium and nitrogen-free medium described by Line and Loutit, the latter medium with sodium thioglycollate, which causes strong reduction conditions in the culture medium itself, is also used in the nitrogen fixation activity assay.
