目的研究辅助治疗期结肠癌湿热证、脾虚证及无症状结肠癌患者血清差异表达的蛋白质。方法收集结肠癌辅助期结肠癌湿热证组、脾虚证组及无症状组患者外周血清各8例,通过双向凝胶电泳以及质谱鉴定和生物信息学分析,寻找在结肠癌辅助治疗期患者血清中特异表达的蛋白质。结果湿热证组较无症状组维生素D结合蛋白升高,血清结合素前体、β-GDP解离抑制因子、载脂蛋白A-I前体和簇蛋白均降低。脾虚证组较无症状组β-GDP解离抑制因子、簇蛋白及血清结合素前体升高,前白蛋白原、载脂蛋白A-I前体及维生素D结合蛋白均降低。结论 6种蛋白差异表达蛋白质的组合性变化可能是辅助治疗期结肠癌的物质基础,具有潜在的诊断、预后标志物或治疗靶点的意义。
Objective To study the differentially expressed proteins of colon cancer with damp - heat syndrome, spleen deficiency syndrome and asymptomatic colon cancer in adjuvant therapy. Methods Collected peripheral blood serum of colon cancer patients with colon-cancer damp-heat syndrome group, spleen deficiency syndrome group and asymptomatic group were collected and analyzed by two-dimensional gel electrophoresis, mass spectrometry identification and bioinformatics analysis. Specific expression of the protein. Results Compared with asymptomatic group, the results showed that vitamin D-binding protein, serum precursor of integrin, β-GDP dissociation inhibitor, apolipoprotein A-I precursor and clusterin were all lower than those without damp-heat syndrome. Compared with asymptomatic group, spleen deficiency syndrome group had higher β-GDP dissociation inhibitor, clusterin and serum precursor, and prealbumin, apolipoprotein A-I precursor and vitamin D-binding protein decreased. Conclusions The combined changes of proteins differentially expressed in 6 proteins may be the material basis for adjuvant therapy of colon cancer with potential diagnostic, prognostic markers or therapeutic targets.
