目的:建立UPLC法测定维生素B_4片中维生素B_4的含量的方法。方法:采用ACQUITY UPLC BEH Shield RP18(100 mm×2.1 mm,1.7μm)色谱柱,流动相为0.02 mol·L~(-1)磷酸二氢钾溶液-甲醇(90:10)(加0.5%的三乙胺,用H_3PO_4调pH至为4.0),流速为0.3 ml·min~(-1),柱温为50℃,检测波长为262 nm。结果:维生素B_4在2.79~139.50μg·ml~(-1)浓度范围内,线性关系良好(r=0.999 8),平均回收率为100.2%,RSD为0.3%(n=9)。结论:本方法简便准确,重复性好,可用维生素B_4片的质量控制。
OBJECTIVE: To establish a method for the determination of vitamin B 4 in vitamin B 4 tablets by UPLC method. METHODS: The mobile phase consisted of 0.02 mol·L -1 potassium dihydrogen phosphate solution - methanol (90:10) with 0.5% (by volume) of ACQUITY UPLC BEH Shield RP18 (100 mm × 2.1 mm, Triethylamine, adjusted to pH 4.0 with H_3PO_4), the flow rate was 0.3 ml · min -1, the column temperature was 50 ℃ and the detection wavelength was 262 nm. Results: The calibration curve of vitamin B 4 showed good linearity (r = 0.999 8) in the range of 2.79-1399.50 μg · ml -1 with an average recovery of 100.2% and a RSD of 0.3% (n = 9). Conclusion: This method is simple and accurate, good reproducibility, available vitamin B_4 tablets quality control.
