磷脂酰胆碱(PC)的生物合成已如前述,其主要合成道路是以胆碱为原料,由ATP提供磷酸形成磷酸胆碱,再由CTP:磷酸胆碱胞苷转移酶(CT)催化生成CDP-胆碱,后者和二脂酰甘油作用最后生成CP。近年来关于CP生物合成的调节已有不少报道,证明CT是CP合成通路中的限速因子,从表1所列各中间代谢物代谢池的大小,可以明显地看出由CT催化的磷酸胆碱转变成CDP-胆碱的反应,是这条合成通路的瓶颈,限制了合成反应的进行。
