目的:研究健肾方含药血清培养成骨细胞移植对骨折愈合过程中血管内皮生长因子(VEGF)mRNA表达的影响。方法:制备4种含药血清,包括健肾方高、低剂量含药血清、福善美含药血清及空白大鼠血清,分别运用4种血清培养乳鼠成骨细胞,并建立骨折模型,将建模后大鼠分5组,分别是模型组、生理盐水组、福善美含药血清合并成骨细胞治疗组、健肾方低剂量含药血清合并成骨细胞治疗组、健肾方高剂量含药血清合并成骨细胞治疗组,治疗采用移植含药血清培养的成骨细胞到骨折部位,用荧光定量PCR方法检测骨折愈合部位VEGFmRNA表达情况。结果:福善美含药血清合并成骨细胞治疗组、健肾方低剂量含药血清合并成骨细胞治疗组、健肾方高剂量含药血清合并成骨细胞治疗组的血管内皮生长因子VEGFmRNA的表达明显增加,与模型组比较差异有统计学意义(P<0.05);健肾方低剂量含药血清合并成骨细胞治疗组与健肾方高剂量含药血清合并成骨细胞治疗组比较差异无统计学意义,但它们与福善美含药血清合并成骨细胞治疗组比较差异有统计学意义(P<0.05)。结论:健肾方含药血清合并成骨细胞移植到骨折部位能显著增加VEGFm-RNA的表达,初步认为其促进骨折愈合的作用与促进VEGFmRNA的表达有关。
Objective: To study the effect of Jianshenfang-containing serum cultured osteoblast transplantation on the expression of vascular endothelial growth factor (VEGF) mRNA during fracture healing. Methods: Four kinds of drug-containing serums were prepared, including high- and low-dose drug-containing serum of Jianshenfang, Fushanmei-containing serum, and blank rat serum. Four types of serum were used to culture neonatal rat osteoblasts, and a fracture model was established. After modeling, the rats were divided into 5 groups: model group, saline group, Fu Shanmei drug-containing serum combined with osteoblast treatment group, Jian Shen Fang low dose drug-containing serum combined with osteoblast treatment group, and Jian Shen Fang high dose. The drug-containing serum was combined with the osteoblasts treatment group. Osteoblasts cultured with transplanted drug-containing serum were used to treat fractures. The expression of VEGF mRNA in fracture healing sites was detected by fluorescence quantitative PCR. RESULTS: The serum level of vascular endothelial growth factor (VEGFmRNA) in the treatment group of Fushanmei combined with serum containing osteoblasts, Jianshenfang low-dosage serum containing osteoblasts, and Jianshenfang high-dose serum containing combined with osteoblasts were compared. Significantly increased expression, compared with the model group, the difference was statistically significant (P <0.05); Jian Shen Fang low dose drug serum combined with osteoblasts treatment group and Jian Shen Fang high dose drug serum combined osteoblast treatment group differences There was no statistical significance, but there was a significant difference between them and the treatment group containing Fushanmei combined with osteoblasts (P<0.05). Conclusion: The serum containing Jianshen prescription combined with osteoblast transplantation to the fracture site can significantly increase the expression of VEGFm-RNA, and it is presumed that its role in promoting fracture healing is related to promoting the expression of VEGF mRNA.
