目的研究血管紧张素转化酶抑制剂(ACEI)对过量饮酒大鼠心脏功能和结构的保护作用。方法 30只大鼠随机分为对照组、酒精组和ACEI组,每组10只。酒精灌胃建立动物模型,并给予ACEI类药物作为保护剂,用超声心动图及超微病理技术观察大鼠心脏功能和结构改变。结果酒精组左室舒张末期内径高于对照组[(6.86±0.64)mmvs(6.29±0.28)mm,P<0.05],收缩末期内径高于对照组[(3.45±0.32)mmvs(2.95±0.25)mm,P<0.05];两组收缩功能参数左室射血分数、每搏输出量、短轴缩短率无统计学差异(P>0.05);舒张功能参数二尖瓣内环舒张早期晚期速度比(Ea/Aa)低于对照组[(0.642±0.126)vs(1.276±0.083),P<0.05]。而ACEI组各参数相对于对照组没有明显改变(P>0.05)。病理形态学观察可见,酒精组线粒体水肿、嵴和膜大部分溶解消失。ACEI组线粒体少部分嵴和膜溶解,损伤程度较酒精组轻。结论 ACEI类药物对过量饮酒大鼠的心肌具有一定的保护作用。
Objective To investigate the protective effect of angiotensin-converting enzyme inhibitor (ACEI) on cardiac function and structure in over-drinking rats. Methods Thirty rats were randomly divided into control group, alcohol group and ACEI group, with 10 rats in each group. The animal models were established by intragastric administration of alcohol and ACEI drugs were given as protective agent. The cardiac function and structure of rats were observed by echocardiography and ultrastructure. Results The diameter of left ventricular end diastolic in alcohol group was significantly higher than that in control group [(6.86 ± 0.64) mm vs (6.29 ± 0.28) mm, P <0.05], and the end systolic diameter was significantly higher in alcohol group than that in control group [(3.45 ± 0.32) mm, P <0.05]. There was no significant difference in systolic function parameters left ventricular ejection fraction, stroke volume per stroke, short axis shortening rate (P> 0.05); diastolic function parameters mitral end systolic early late velocity The ratio (Ea / Aa) was lower than that of the control group [(0.642 ± 0.126) vs (1.276 ± 0.083, P <0.05]. The ACEI group parameters relative to the control group did not change significantly (P> 0.05). Histopathological examination revealed that most of mitochondria edema, crest and membrane in alcohol group disappeared. ACEI group mitochondria a small part of the crest and membrane dissolution, the degree of injury lighter than the alcohol group. Conclusion ACEI drugs have a protective effect on myocardium of over-drinking rats.
