目的探讨光动力治疗对胶质瘤细胞表面侵袭性特异结构的影响。方法应用细胞培养及原子力显微镜技术,对5-氨基乙酰丙酸(5-ALA)作用前后U251胶质瘤细胞表面特异膜结构进行纳米尺度形态学对比研究。结果胶质瘤细胞膜表面大量伪足结构和微绒毛结构经5-ALA治疗,由直径约400nm微绒毛结构,呈指状突起,长度1μm,改变为直径300nm球状颗粒,指状突起消失,膜状伪足、丝状伪足消失。结论经5-ALA介导的光动力治疗对胶质瘤细胞侵袭性具有显著的破坏作用。
Objective To investigate the effects of photodynamic therapy on the invasive glioma cell surface specific structure. Methods Cell culture and atomic force microscopy were used to compare the surface-specific membrane structures of U251 glioma cells before and after 5-aminolevulinic acid (5-ALA) treatment with nanoscale morphology. Results A large number of pseudopod structures and microvilli structures on the surface of glioma cells were treated with 5-ALA. The micro-villi structures were composed of micro-villi with a diameter of about 400 nm. The length was 1 μm and the diameter was about 300 nm. The finger- Pseudopodia, filopodia disappear. Conclusion 5-ALA-mediated photodynamic therapy has a significant impact on invasiveness of glioma cells.
