目的:探讨重组腺病毒Rb94(Ad-Rb94)基因转染对宫颈癌细胞辐射敏感性的影响。方法:将Ad-Rb94基因转染宫颈癌细胞株Hela,进行137Csγ射线照射,数字随机表法分为空白对照组、Ad-LacZ组、Ad-Rb94组、照射组和Ad-Rb94联合照射组。用RT-PCR法和免疫荧光法检测Rb-94基因mRNA和蛋白水平的表达,以MTT法观察细胞的生长曲线和单细胞凝胶电泳(SCGE)实验观察细胞DNA的损伤。结果:RT-PCR法和免疫荧光法检测到在Hela细胞有Rb-94基因的表达,Ad-Rb94组、照射组和Ad-Rb94联合照射组Hela细胞的生长均受到抑制,其中联合照射组的抑制效应最强(P<0.05)。照射组和联合照射组细胞照射后出现DNA损伤,联合组损伤程度强于照射组,差异有统计学意义(P<0.05)。结论:Ad-Rb94基因联合照射能明显增强人宫颈癌细胞的辐射敏感性。
Objective: To investigate the effect of recombinant adenovirus Rb94 (Ad-Rb94) gene transfection on the radiosensitivity of cervical cancer cells. Methods: Ad-Rb94 gene was transfected into cervical cancer cell line Hela and irradiated with 137Csγ ray. The cells were divided into blank control group, Ad-LacZ group, Ad-Rb94 group, irradiation group and Ad-Rb94 combined irradiation group. The expression of Rb-94 mRNA and protein was detected by RT-PCR and immunofluorescence. The cell growth curve and single cell gel electrophoresis (SCGE) were used to observe the DNA damage by MTT assay. Results: The expression of Rb-94 gene in Hela cells was detected by RT-PCR and immunofluorescence. The growth of Hela cells was inhibited in Ad-Rb94 group, irradiation group and Ad-Rb94 combined irradiation group, and the combined irradiation group The strongest inhibitory effect (P <0.05). DNA damage occurred in the irradiation group and the combined irradiation group, and the damage degree in the combined group was stronger than that in the irradiation group (P <0.05). Conclusion: Ad-Rb94 gene combined with irradiation can significantly enhance the radiation sensitivity of human cervical cancer cells.
