目的观察单味关木通、龙胆泻肝汤和去当归、生地、甘草的龙胆泻肝汤给药后大鼠肾功能和组织形态学变化。方法大鼠随机分为正常对照组、单味关木通组、龙胆泻肝汤组和龙胆泻肝汤去归地草方组,正常对照组给予等量蒸馏水,后3组分别按6、17.5、14.5 g/kg给以相应方药的水煎液灌胃,各组关木通含量均为6 g/kg,连续8周。给药结束后,测定大鼠血、尿生化指标,观察肾脏组织形态学改变。结果各水煎液灌胃组,均有明显肾损伤表现,其中以单味关木通组损伤最为严重,龙胆泻肝汤组较轻,去归地草的龙胆泻肝汤组较龙胆泻肝汤组损伤重,个别指标甚至高于单味关木通组。结论实验剂量的关木通与配伍关木通的两个复方均可引起肾损伤,龙胆泻肝汤去当归、生地、甘草方的肾毒性较龙胆泻肝汤强。提示含关木通配伍的复方的毒性作用不等于关木通,龙胆泻肝汤方中当归、生地、甘草的配伍对关木通的肾损伤有一定的减毒作用。
Objective To observe the changes of renal function and histomorphology in rats after the treatment of Longdan Xiegang Decoction with Guanweitong, Longdanxiegan Decoction, and Angelica, Rehmannia, and Glycyrrhiza. Methods Rats were randomly divided into normal control group, Shanwei Guan Mu Tong group, Long Dan Xie Gan Decoction group, and Long Dan Xiegang Tang De Gui Di Cao Fang group. The normal control group was given the same amount of distilled water, and the latter three groups were treated as 6, 17.5. 14.5 g/kg was given to the corresponding decoction of decoction. The content of Guanmutong in each group was 6 g/kg for 8 weeks. After the administration, blood and urine biochemical indicators were measured and the morphological changes of the kidneys were observed. Results The rats in each group had obvious renal injury, among which the single-dose Guanmutong group had the most serious damage, and the Longdanxiegan decoction group was lighter. The Longdanxiegan decoction group was better than the Longxigong diarrhea group. Liver soup group was heavily damaged, and individual indicators were even higher than the single-flavor Guanmutong group. Conclusion Both the experimental dose of Guan Mu Tong and the combination of Guan Mu Tong can cause kidney injury. Long Dan Xie Gan Decoction to Angelica, Rehmannia and Glycyrrhiza Herb has stronger renal toxicity than Long Dan Xie Gan Tang. It is suggested that the toxic effects of the compounds containing Guanmutong are not equal to Guan Mu Tong, and the compatibility of Angelica Root, Radix Rehmanniae, and Radix Glycyrrhizae in the Long Dan Xie Tang Decoction has a certain attenuating effect on the renal damage of Guan Mu Tong.
