目的研究毛蒟水提物对CCl_4所致小鼠急性肝损伤的保护作用及其机制。方法雄性昆明种小鼠随机分为空白对照组,模型组,毛蒟水提物低、高剂量组,采用CCl_4建立急性肝损伤模型。观察毛蒟水提物对模型组小鼠血清中天冬氨酸转氨酶(AST)、丙氨酸氨基转移酶(ALT)活性,小鼠肝脏指数和肝组织中Egr-1、TNF-αmRNA表达的影响,以及肝组织的病理学改变。结果毛蒟水提物给药组可明显降低CCl_4所致肝损伤小鼠血清中AST、ALT的活性(P<0.01),降低肝脏组织中Egr-1和TNF-αmRNA的表达水平(P<0.05);可明显改善CCl_4所致肝脏病理细胞损伤,炎症细胞明显减少,且高剂量组更为显著。结论毛蒟水提物对CCl_4所致小鼠急性肝损伤具有明显的保护作用,其机制可能与抑制Egr-1和TNF-α等炎症通路因子的表达有关。
Objective To study the protective effect and the possible mechanism of Muddy water extract on acute liver injury induced by CCl_4 in mice. Methods Male Kunming mice were randomly divided into blank control group, model group, low and high dose of Muddy water extract, and acute liver injury model was established by CCl_4. To observe the effect of Mudan water extract on serum AST, ALT activity, liver index and Egr-1, TNF-αmRNA expression in model group Impact, and pathological changes in liver tissue. Results The Mudan water extract group could significantly reduce the activity of AST and ALT (P <0.01) and reduce the expression of Egr-1 and TNF-αmRNA (P <0.05) in the liver of CCl 4 -induced liver injury ); Can significantly improve the CCl_4-induced liver pathological cell injury, inflammatory cells decreased significantly, and high-dose group was more significant. Conclusion The aqueous extract of catfish has a protective effect on CCl_4-induced acute liver injury in mice. The mechanism may be related to the inhibition of the expression of inflammatory factors such as Egr-1 and TNF-α.
