目的探讨椰子多肽提取液的分离提取方法,找出椰子多肽提取液最佳的提取方法。方法取椰肉,进行双酶水解,分离出清液,清液进行吸附分离纯化,再用滤膜进行超滤,即得椰子提取物浓缩液。结果椰子多肽提取液最佳提取参数为:酶解温度为45℃、胃蛋白酶的酶解时间为8h、胰酶的酶解时间为3h,胃蛋白酶的酶解pH值为1.5~1.7,胰酶的酶解pH值为7.5~7.7,两种酶的用量都为椰肉重量的1.5%,料液比1:10,提取所得的椰子多肽提取液经测定其多肽含量为15.8mg/mL。结论试验结果表明,椰子多肽的分离提取工艺是可行的。
Objective To explore the method of separating and extracting coconut peptide extract and to find out the best extraction method of coconut peptide extract. Method Take coconut meat, double enzyme hydrolysis, separation of the supernatant, the supernatant was purified by adsorption, and then the filter membrane ultrafiltration, that coconut extract concentrate. Results The optimal extraction parameters of coconut peptide extract were: enzymolysis temperature 45 ℃, pepsin digestion time 8h, trypsin digestion time 3h, pepsin pH 1.5 ~ 1.7, trypsin The enzymolysis pH was 7.5-7.7. The amount of both enzymes was 1.5% of the weight of copra, the ratio of material to liquid was 1:10. The extracted coconut peptide extract was determined to have a polypeptide content of 15.8 mg / mL. Conclusion The experimental results show that the coconut peptide separation and extraction process is feasible.
