目的研究信号肽在真核表达系统中对金属蛋白酶多抗原表位DNA疫苗诱导小鼠免疫应答影响。方法人工合成含有金属蛋白酶多个抗原表位的DNA序列,将连接信号肽与不连接信号肽的该序列分别连接pIRESneo表达载体,构建的2种质粒免疫小鼠,ELISA法、出血中和法检测免疫后效价。结果 2种质粒诱导小鼠产生的抗体效价10-2为阳性,均能有效减轻金属蛋白酶引起的出血。结论信号肽的有无对金属蛋白酶DNA疫苗在小鼠体内表达没有影响。
Objective To study the effect of signal peptide on immune response induced by metalloproteinase polyepitope DNA vaccine in mouse eukaryotic expression system. Methods The DNA sequence containing multiple epitopes of metalloproteinase was synthesized. The pIRESneo expression vector was constructed by ligating the signal peptide with the signal peptide without signal peptide. The two plasmids were immunized with ELISA, and the method of hemorrhage neutralization Post-immune titer. Results The results showed that the antibody titer 10-2 produced by the two plasmids in mice was positive, and all of them could effectively reduce the metalloproteinase - induced bleeding. Conclusion The presence of signal peptide has no effect on the expression of metalloproteinase DNA vaccine in mice.
