目的探讨叶酸对人脂肪间充质干细胞(hADSCs)成脂分化过程的影响及机制。方法以正常叶酸(Control)和无叶酸(FD)成脂诱导液对hADSCs细胞进行成脂诱导,不同时间分别采用油红O染色观察细胞内脂滴生成情况;采用SYBR Green实时荧光定量RT-PCR方法检测成脂分化相关转录调控基因PPARγ和C/EBPα的表达;采用蛋白质印迹法检测PPARγ蛋白的表达。结果与正常叶酸组相比,无叶酸组脂滴数量明显减少;两组细胞成脂分化相关转录调控基因PPARγ和C/EBPαmRNA表达无明显差异;PPARγ蛋白表达明显低于正常叶酸组。结论叶酸可通过上调PPARγ的表达而促进hADSCs细胞成脂分化。
Objective To investigate the effect of folic acid on adipogenic differentiation of human adipose-derived mesenchymal stem cells (hADSCs) and its mechanism. Methods The hADSCs cells were induced to adipogenesis by normal adipose-induced and folate-free adipogenic medium. The lipid droplets were observed by oil red O staining at different time points. The expression of lipid droplets was detected by SYBR Green real-time quantitative RT-PCR Methods The expression of PPARγ and C / EBPα related to adipogenic differentiation were detected. The expression of PPARγ protein was detected by Western blotting. Results Compared with the normal folic acid group, the number of lipid droplets in the group without folic acid was significantly reduced. There was no significant difference in PPARγ and C / EBPα mRNA expression between the two groups. The PPARγ protein expression was significantly lower than that in the normal folic acid group. Conclusion Folic acid can promote the adipogenic differentiation of hADSCs by up-regulating the expression of PPARγ.
