目的研究丹红注射液对氯化锂-匹罗卡品点燃慢性颞叶癫痫(temporal lobe epilepsy,TLE)大鼠海马神经元及GFAP表达的干预作用。方法建立氯化锂-匹罗卡品点燃颞叶癫痫大鼠模型,将50只造模成功的颞叶癫痫大鼠随机分为颞叶癫痫组(生理盐水10ml/kg腹腔注射)及丹红治疗组(丹红注射液10ml/kg腹腔注射),另选10只大鼠作为正常对照组;每组大鼠随机分为5个不同时间点:24h、3d、7d、15d及30d。采用尼氏染色及免疫组化染色,检测每组大鼠不同时间点海马CA1区神经元存活数目及GFAP表达。结果与正常对照组相比,颞叶癫痫组大鼠海马CA1区存活神经元数目明显减少,海马GFAP表达显著增多(P<0.05);与颞叶癫痫组相比,丹红治疗组大鼠海马CA1区存活神经元数目明显增多,海马GFAP表达显著减少(P<0.05)。结论丹红注射液可以减轻颞叶癫痫大鼠海马神经元脱失及减少GFAP表达,有助于减轻大鼠发作程度及阻断慢性颞叶癫痫的形成和发展。
Objective To investigate the effects of Danhong injection on hippocampal neurons and GFAP expression in lithium chloride-pilocarpine-induced chronic temporal lobe epilepsy (TLE) rats. Methods The models of temporal lobe epilepsy induced by lithium chloride-pilocarpine were established. Fifty temporal lobe epilepsy rats with successful modeling were randomly divided into temporal lobe epilepsy group (intraperitoneal injection of saline 10ml / kg) and Danhong (Danhong injection 10ml / kg intraperitoneal injection), and another 10 rats as normal control group; each group of rats were randomly divided into five different time points: 24h, 3d, 7d, 15d and 30d. Nissl staining and immunohistochemistry were used to detect the number of neurons and the expression of GFAP in hippocampal CA1 area of rats at different time points. Results Compared with the normal control group, the number of neurons in the hippocampal CA1 subfield in the temporal lobe epilepsy group was significantly decreased and the GFAP expression was significantly increased in the hippocampus (P <0.05). Compared with the temporal lobe epilepsy group, the hippocampus There was a significant increase in the number of neurons in CA1 area, and a significant decrease in GFAP expression in hippocampus (P <0.05). Conclusion Danhong injection can reduce the loss of hippocampal neurons and decrease the expression of GFAP in temporal lobe epilepsy rats, which may help to reduce the degree of seizures and block the formation and development of chronic temporal lobe epilepsy.
