雷公藤多甙(GTW)是从卫茅科植物雷公藤中提取的一种中成药,它对成年男子,雄性大鼠和小鼠都有明确的抗生育作用。在GTW进一步分离过程中得到一个单体化合物、雷藤氯内酯醇(T_4)。经实验室观察,雄性大鼠每天灌服50μg/kg,连服6周即可达不育。其抗生育效价为GTW的200倍,毒副作用明显下降。此剂量也不影响大鼠血清中睾酮和LH水平。但在离体的间质细胞培养过程中加入T_4,则发现睾酮分泌量明显下降。认为T_4对间质细胞功能有直接抑制作用。而T_4对垂体细胞分泌LH功能是否有直接影响尚需进一步探讨。本研究观察了T_4对在体及离体垂体细胞分泌与贮藏LH功能的影响。
Tripterygium wilfordii (GTW) is a proprietary Chinese medicine extracted from Tripterygium wilfordii. It has clear anti-fertility effects in adult males, male rats and mice. In the further separation of GTW, a monomeric compound, Letolactone (T_4) was obtained. According to laboratory observations, male rats were given 50 μg/kg daily for 6 weeks for infertility. Its anti-fertility price is 200 times that of GTW, and its toxic and side effects have dropped significantly. This dose also does not affect testosterone and LH levels in rat serum. However, when T_4 was added during the culture of isolated mesenchymal cells, the secretion of testosterone was significantly reduced. It is believed that T_4 has a direct inhibitory effect on the function of mesenchymal cells. However, whether T4 has a direct effect on the secretion of LH from pituitary cells still needs further investigation. This study investigated the effects of T_4 on the secretion and storage of LH in vivo and in vitro.
