目的探索尿路清对解脲支原体(Uu)感染的体外抗菌药效,为中医药治疗本病提供依据。方法63个Uu临床分离株采用肉汤稀释法判定耐红霉素和四环素(MIC≥16μg/mL,参照美国国家临床实验室标准委员会1995年标准);以Uu1~14型抗血清,采用MIT法作血清分型后,经3次传代成纯培养物后接种于96孔板小孔中,药液倍比稀释成250mg/mL~0.48mg/mL,37℃培养,加药组72h后仍不出现培养基由黄色变为红色的最小药物浓度为该药的最小抑菌浓度(MIC)。结果尿路清对63株Uu临床株MIC范围为0.48~15.63mg/mL,MIC50≤1.95mg/mL,MIC90≤3.91mg/mL。Uu临床株对尿路清的敏感性比对四环素高得多(χ2=18.38,P<0.01);Uu临床株对尿路清的敏感性与对红霉素的敏感性之间无明显联系(χ2=0.29,P>0.05)。14种和未定型Uu血清型别中,除11,1,2,3型对尿路清的敏感性稍差(极少数菌株MIC≥7.81mg/mL),其余血清型别对尿路清的敏感性均较高(MIC≤3.91mg/mL)。结论中药尿路清对63株不同血清型、耐药(四环素、红霉素)菌株Uu显示出了较强的体外抑菌作用。
Objective To explore the antibacterial efficacy of urinary tract clearing Ureaplasma urealyticum (Uu) infection in vitro and provide basis for the treatment of this disease by Chinese medicine. Methods 63 Uu clinical isolates were determined by broth dilution method to determine erythromycin-resistant and tetracycline (MIC ≥ 16 μg/mL, refer to the 1995 standards of the National Clinical Laboratory Standards Committee of the United States); Uu1~14 antiserum and MIT method were used. After serotyping, after three passages into pure cultures, they were inoculated into 96-well plate wells and diluted to 250 mg/mL to 0.48 mg/mL, and incubated at 37°C. The minimum drug concentration at which the medium changes from yellow to red is the minimum inhibitory concentration (MIC) of the drug. Results The MIC of the 63 Uu clinical isolates from urinary tract was from 0.48 to 15.63 mg/mL, MIC50≤1.95 mg/mL, and MIC90≤3.91 mg/mL. The sensitivity of Uu clinical strains to urinary tract clearance was much higher than that of tetracycline (χ2=18.38, P<0.01); there was no clear relationship between the sensitivity of Uu clinical strains to urinary tract clearance and the sensitivity to erythromycin ( Χ2=0.29, P>0.05). Among the 14 U(superscript u) and unidentified Uu serotypes, the susceptibility to urinary tract clearance was slightly lower than those of U1 serotypes (very small strain MIC ≥ 7.81 mg/mL). The remaining serotypes were not sensitive to urinary tract urine clearance. Sensitivity is higher (MIC ≤ 3.91 mg/mL). Conclusion Urealuqing showed strong antibacterial effect in vitro on 63 strains of Uu, which had different serotypes and resistance (tetracycline, erythromycin).
