介绍了一种新的测定半胱氨酸和还原型谷胱甘肽的荧光光度法。 N- [对 - (2 -苯并口恶唑基 ) -苯基 ]-马来酰亚胺是一种有很弱荧光的活性试剂 ,可以和硫醇定量反应 ,其加成产物有强烈荧光。利用加成产物的荧光 ,可以测定半胱氨酸和还原型谷胱甘肽 ,检出限量分别是 4 .0× 10 - 1 0 m ol· L- 1 和 3.2× 10 - 1 0 m ol· L- 1 (SNR=3)。该方法可以用于蛋白质和胱氨酸水解液中半胱氨酸的测定。
A new fluorimetric method for the determination of cysteine and reduced glutathione was introduced. N- [p- (2-benzoxazolyl) -phenyl] -maleimide is a weakly fluorescent reagent that can react quantitatively with thiols and its addition product has strong fluorescence. Cysteine and reduced glutathione can be measured by the fluorescence of the adduct, with detection limits of 4.0 × 10 -1 mol -1 and 3.2 × 10 -1 mol -1 respectively L-1 (SNR = 3). This method can be used for the determination of cysteine in protein and cystine hydrolyzate.
