本研究采用组织培养、高效液相色谱等方法,确定了羊草的休眠类型为抑制物引起的生理性休眠,主要抑制部位是胚乳,关键抑制物之一是脱落酸(ABA);羊草种子的休眠期为4年左右,5至6年达到自然发芽率高峰,此后发芽率开始下降;外源植物激素CAS和6─BA对解除羊草种子的休眠具有促进作用。贮存1年的深度休眠的羊草种子,用低温流水浸泡法处理后能使自然发芽率(土培法,35.0%)提高到88.5%(土培法).不同发芽床上进行的发芽率比较试验表明,土培法和沙培法明显优于纸培法,以土培法的发芽率为最高。
In this study, tissue culture and high performance liquid chromatography (HPLC) were used to determine the dormancy types of Leymus chinensis. The main inhibitory sites were endosperm. One of the key inhibitors was abscisic acid (ABA) The dormant period of about 4 years, 5 to 6 years to reach the peak of natural germination, and germination rate began to decline; exogenous plant hormones CAS and 6 ─ BA to relieve the dormancy of Leymus chinensis seed has a role in promoting. Deep-dormant Leymus chinensis seeds stored for one year increased the rate of natural germination (soil culture, 35.0%) to 88.5% (soil culture) after being soaked in low temperature water. The germination rate of different germination bed comparison test showed that soil culture and sand culture method was significantly better than the paper culture method to soil culture method for the highest germination rate.
