目的水合氯醛对小鼠生长发育、学习记忆和治疗指数的影响。方法用各组小鼠体重变化趋势观察水合氯醛对小鼠生长发育的影响,避暗法和跳台法观察水合氯醛对小鼠学习记忆的影响,用分组法测量水合氯醛对小鼠催眠的ED50、LD50和治疗指数。结果在跳台和避暗实验中,与NS组相比,第一天C1、C2、C3(水合氯醛剂量分别为0.005、0.0075、0.01ml·10g-1)潜伏期缩短(P<0.01),避暗实验C1、C3组和跳台实验C2、C3组错误次数增加(P<0.01),第二天,与NS组相比,避暗实验中C1、C3组错误次数增加(P<0.05或P<0.01),C1组潜伏期缩短(P<0.05),第三、四天未见显著性差异(P>0.05);各组小鼠在1-5d体重大致呈增长趋势,组间比较无明显差异。结论水合氯醛可在短期内损害小鼠学习记忆能力,对生长发育无明显影响,治疗指数为3.842,用药安全范围较小。
Objective Chloral hydrate on mouse growth and development, learning and memory and therapeutic index. Methods The body weight of each group was used to observe the effects of chloral hydrate on the growth and development of mice. The effects of chloral hydrate on the learning and memory of mice were observed by the method of avoiding darkness and jumping. The hypnotic effects of chloral hydrate ED50, LD50 and therapeutic index. Results Compared with the NS group, the latency of C1, C2 and C3 (0.005, 0.0075 and 0.01 ml · 10g-1, respectively) on the first day was shorter (P <0.01) In the dark test, the number of errors in C1, C3 group was significantly increased (P <0.05 or P < 0.01). The incubation period in C1 group was shorter than that in C1 group (P <0.05). There was no significant difference in the third and fourth days (P> 0.05). Conclusion Chloral hydrate can impair the ability of learning and memory in short term and has no obvious effect on the growth and development of mice. The therapeutic index is 3.842, and the medication safety range is small.
