目的:观察吗啡对胸腺细胞亚群DNA含量和增殖指数的影响及人参四物汤对其调节作用。方法:采用小剂量递增给药建立吗啡成瘾动物模型后,经流式细胞仪分析各组小鼠胸腺CD4、CD8亚群细胞DNA含量及增殖指数的情况。结果:吗啡成瘾组小鼠CD4、CD8亚群细胞S期、G2+M期和增殖指数显著低于正常组和人参四物汤治疗组动物,而G0/G1期(静止期)的DNA含量明显增高,提示吗啡使胸腺细胞DNA复制受阻导致功能细胞减少。人参四物汤可对抗吗啡的上述作用,促进免疫细胞DNA合成、细胞增殖分化。
OBJECTIVE: To observe the effect of morphine on DNA content and proliferation index of thymocyte subpopulation and the regulatory effect of Ginseng Siwu Decoction on it. METHODS: After establishing an animal model of morphine addiction with low-dose incremental administration, the DNA content and proliferation index of CD4 and CD8 subpopulation cells in the thymus of mice in each group were analyzed by flow cytometry. RESULTS: The levels of S phase, G2+M phase and proliferation index of CD4 and CD8 subpopulations in morphine addiction group were significantly lower than those in normal group and Ginseng Siwutang treatment group, while the DNA content in G0/G1 phase (stationary phase) was significantly increased. , suggesting that morphine makes thymus cell DNA replication is blocked resulting in a decrease in functional cells. Ginseng Siwu Decoction can fight the above-mentioned effects of morphine and promote DNA synthesis and cell proliferation and differentiation in immune cells.
