目的:观察不同浓度的鸡肉、鸭肉提取液分别对体外培养人肺腺癌A549细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)及DNA合成的干预作用,探讨鸡肉、鸭肉提取液对体外培养人肺腺癌A549细胞影响的作用机制,以期为临床肿瘤患者合理饮食提供指导。方法:体外培养人肺腺癌A549细胞株,细胞生长至对数生长期后,随机分为9组进行培养。采用VEGF-ELISA法检测肺腺癌A549细胞上清液中的VEGF浓度,BrdU-ELISA法检测肺腺癌A549细胞DNA的合成情况。结果:与对照组相比,鸡肉提取液、鸭肉提取液均能促进肺腺癌A549细胞血管内皮生长因子的生长,无统计学差异(P>0.05)。鸡肉提取液、鸭肉提取液均能够促进肺腺癌A549细胞DNA的合成,差异有统计学意义(P<0.01),且随着剂量的增加DNA的合成越多。结论:鸡肉、鸭肉提取液并没有通过促进肺腺癌A549血管内皮生长因子生长来促进肿瘤细胞生长。鸡肉、鸭肉提取液均能够促进肺腺癌A549细胞DNA的合成,使细胞的DNA合成期(S Phase;S期)增多,S期增多可能是其促进细胞增殖的机制之一。
OBJECTIVE: To observe the intervention effects of different concentrations of chicken and duck extract on vascular endothelial growth factor (VEGF) and DNA synthesis in cultured human lung adenocarcinoma A549 cells in vitro and to explore the effect of chicken and duck extract on vitro Culture human lung adenocarcinoma A549 cells affect the mechanism of action in order to provide a reasonable guide for clinical cancer patients. Methods: Human lung adenocarcinoma A549 cells were cultured in vitro. After the cells grew to logarithmic growth phase, they were randomly divided into 9 groups and cultured. The concentration of VEGF in lung adenocarcinoma A549 cells supernatant was detected by VEGF-ELISA and the DNA synthesis in lung adenocarcinoma A549 cells was detected by BrdU-ELISA. Results: Compared with the control group, chicken extract and duck extract could promote the growth of vascular endothelial growth factor in A549 cells. There was no significant difference (P> 0.05). Chicken extract and duck extract all could promote the DNA synthesis of lung adenocarcinoma A549 cells, the difference was statistically significant (P <0.01), and the more DNA was synthesized with the dose increasing. Conclusion: Chicken and duck extract did not promote the growth of tumor cells by promoting the growth of human lung adenocarcinoma A549 vascular endothelial growth factor. Chicken and duck extract could promote the DNA synthesis of lung adenocarcinoma A549 cells and increase the DNA synthesis phase (S Phase; S phase) of cells. Increasing S phase may be one of the mechanisms of cell proliferation.
