在含不同浓度NaCl的MS培养基上,继代5次培养大豆发育子叶的愈伤组织,分析了不同盐浓度培养的大豆愈伤组织的蛋白质、氨基酸和可溶性糖的含量。经测定表明,盐胁迫下,大豆愈伤组织的蛋白质,总氨基酸含量都比对照高,可溶性糖含量比对照低。NaCl浓度在0~0.1%范围内,随NaCl浓度的增加,蛋白质与总氨基酸的含量逐渐增加,当NaCl浓度增至0.15%时,蛋白质和总氨基酸含量又降低。在含NaCl0.07%培养基中,大豆愈伤组织的脯氨酸含量最高,为对照的2.72倍。盐胁迫下、脯氨酸、丙氨酸、精氨酸和组氨酸的含量变化与总氨基酸含量变化不一致。
The callus of soybean was cultured on MS medium containing different concentrations of NaCl for 5 times, and the contents of protein, amino acid and soluble sugar in soybean callus cultured in different salt concentrations were analyzed. The results showed that under salt stress, the contents of protein and total amino acids in soybean callus were higher than those in the control, and the content of soluble sugar was lower than the control. The concentration of NaCl and NaCl in the range of 0-0.1% increased with the increase of NaCl concentration. The content of protein and total amino acid increased when NaCl concentration increased to 0.15%. In 0.07% NaCl medium, the content of proline in soybean callus was the highest, 2.72 folds of the control. Under salt stress, the content of proline, alanine, arginine and histidine changed inconsistent with the total amino acid content.
