目的观察DNA甲基化转移酶1(DNMT1)在非小细胞肺癌(NSCLC)组织、癌旁组织及正常肺组织中的表达,通过其表达水平的分析,探讨其与临床特征的关系。方法选取睢宁县人民医院胸外科手术切除的肺癌标本,分别取癌组织54例,癌旁组织48例,另取周边正常组织24例为对照组,行免疫组化检测,分析各组织中DNMT1表达差异及与临床各指标之间关系。结果 DNMT1在NSCLC的癌组织、癌旁组织、正常肺组织中阳性表达率分别39/54(72.2%)、30/48(62.5%)、0/24(0%),与正常肺组织比较差异有统计学意义(P<0.01)。DNMT1的表达与NSCLC组织的分化程度、TNM分期、患者的年龄、性别、吸烟、病理类型无相关性。结论 DNMT1在NSCLC癌组织、癌旁组织中出现高表达,在正常肺组织中表达缺失,提示DNMT1参与NSCLC发病机制,可以作为病理诊断NSCLC的标志物。DNMT1与NSCLC的临床特征之间无相关性,不能作为预后的指标。
Objective To investigate the expression of DNA methyltransferase 1 (DNMT1) in non-small cell lung cancer (NSCLC) tissues, paracancerous tissues, and normal lung tissues. Through the analysis of the expression level, the relationship between DNA methylation transferase 1 (DNMT1) and clinical characteristics was explored. Methods Lung cancer specimens were collected from Thoracic Surgery Department of People’s Hospital of Suining County. 54 cases of cancer tissues and 48 cases of paracancerous tissues were collected. 24 cases of normal tissues were used as control group. Immunohistochemistry was performed to analyze the expression of DNMT1 in each tissue. Differences and relationships with clinical indicators. Results The positive rates of DNMT1 in NSCLC tissues were 39/54 (72.2%), 30/48 (62.5%), and 0/24 (0%), respectively, which were different from normal lung tissues. Statistically significant (P<0.01). The expression of DNMT1 was not associated with the degree of differentiation of NSCLC tissues, TNM staging, age, sex, smoking, and pathological type of patients. Conclusions DNMT1 is highly expressed in NSCLC tissues and adjacent tissues, and is absent in normal lung tissues. This suggests that DNMT1 may be involved in the pathogenesis of NSCLC and may be used as a marker for the pathological diagnosis of NSCLC. There was no correlation between the clinical features of DNMT1 and NSCLC and could not be used as an indicator of prognosis.
