目的观察脓毒症大鼠心肌高迁移率族蛋白B1(HMGB1)、血清肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)、肌钙蛋白Ⅰ(TnⅠ)的水平,探讨桃红芩连汤治疗脓毒症心肌损伤机制。方法 48只清洁级健康雄性Wistar大鼠随机分成假手术组(Sham组)、脓毒症模型组(CLP组)、脓毒症中药治疗组(ZY组),每组16只。于手术后24、48h分别检测TNF-α、IL-6、TnⅠ、HMGB1浓度,光镜观察心肌病理变化。结果与Sham组比较,CLP组在24、48hTNF-α、IL-6、TnⅠ及HMGB1水平均显著升高,差异有统计学意义(P<0.01);与CLP组同期比较,ZY组24、48h时TNF-α、IL-6、TnⅠ及HMGB1浓度均明显下降,差异亦有统计学意义(P<0.05)。光镜下CLP组和ZY组可见心肌损伤,CLP组较ZY组损伤明显。结论桃红芩连汤可降低脓毒症相关炎症因子水平,对心肌有保护作用。
Objective To observe the levels of high mobility group box protein 1 (HMGB1), serum tumor necrosis factor α (TNF-α), interleukin-6 (IL-6) and troponin Ⅰ (TnⅠ) in septic rats, To explore the mechanism of Taohong Qinlian Decoction in treating myocardial injury in sepsis. Methods Forty - eight healthy male Wistar rats were randomly divided into sham operation group (Sham group), sepsis model group (CLP group) and sepsis Chinese medicine treatment group (ZY group), with 16 rats in each group. The concentrations of TNF-α, IL-6, TnⅠ and HMGB1 were detected at 24 and 48 hours after operation. The pathological changes of myocardium were observed by light microscope. Results Compared with Sham group, the levels of TNF-α, IL-6, TnⅠ and HMGB1 in CLP group were significantly increased at 24 and 48 hours (P <0.01). Compared with CLP group, The concentrations of TNF-α, IL-6, TnⅠ and HMGB1 were significantly decreased, the difference was also statistically significant (P <0.05). Under light microscope, myocardial injury was observed in CLP group and ZY group, while injury in CLP group was more obvious than that in ZY group. Conclusions Taoqinlianlian Decoction can reduce the level of sepsis-related inflammatory cytokines and have protective effect on myocardium.
