将人TNFα基因克隆到重组逆转录病毒载体pRUFneo,构建了含人TNF基因重组逆转录病毒载体RUF-TNF。将RUF-TNF转染包装细胞FlyA13,经药物筛选获得稳定分泌含人TNF基因重组逆转录病毒的细胞株FlyRUFN14。FlyRUFN14产生的病毒上清液能直接转染靶细胞Saos-2。本研究建立的逆转录病毒介导的TNF基因转移系统为肝癌及其他恶性肿瘤的基因治疗提供了条件。
The human TNFα gene was cloned into the recombinant retroviral vector pRUFneo to construct a recombinant retrovirus vector RUF-TNF containing human TNF gene. RUF-TNF was transfected into the packaging cell FlyA13, and the cell line FlyRUFN14 stably secreting the recombinant retrovirus containing the human TNF gene was obtained through drug screening. The virus supernatant produced by FlyRUFN14 can directly transfect the target cell Saos-2. The retroviral-mediated TNF gene transfer system established in this study provides conditions for the gene therapy of liver cancer and other malignant tumors.
