目的建立有一定免疫功能的B细胞淋巴瘤小鼠模型。方法 38只BALB/c小鼠,其中32只小鼠尾静脉注射1×106个A20细胞后分为实验组16只和对照组16只,分别腹腔注射姥鲛烷0.5mL和生理盐水0.5mL,观察2组小鼠成瘤情况、时间、部位和成瘤率,并分别取各脏器组织行组织病理检查,采用免疫组织化学法检测肿瘤组织免疫表型。另6只小鼠作为空白对照组,采用流式细胞术检测3组小鼠的免疫功能。结果对照组小鼠成瘤率为43.8%,成瘤时间为(72.6±8.7)d;实验组小鼠成瘤率为87.5%,成瘤时间为(32.5±4.1)d,差异有统计学意义(P<0.05);荷瘤鼠的免疫功能未明显改变,实验组成瘤小鼠CD19阳性细胞表达率为(90.01±3.51)%,对照组为(87.79±5.62)%,差异无统计学意义(P>0.05);实验组、对照组小鼠第30天NK细胞数(40.7±2.2和60.3±1.9)均明显高于空白对照组(15.1±0.6)(P<0.05),对照组高于实验组(P<0.05)。结论姥鲛烷联合A20细胞株成功构建了有一定免疫功能的播散性B细胞淋巴瘤小鼠动物模型,成瘤率高,成瘤时间短,且保留了较好的免疫功能。
Objective To establish a mouse model of B cell lymphoma with certain immune function. Methods Thirty-eight BALB / c mice were randomly divided into experimental group (16 rats) and control group (16 mice). Thirty-two mice were injected intravenously with 0.5 mL pristane and 0.5 mL normal saline respectively. The tumorigenicity, time, location and tumor formation rate of the two groups of mice were observed. The histopathological examination of each organ was performed. Immunohistochemistry was used to detect the immunophenotype of tumor. The other 6 mice served as blank control group, and the immune function of 3 groups of mice was detected by flow cytometry. Results The tumor formation rate was 43.8% in the control group and the tumorigenic time was (72.6 ± 8.7) days. The tumor formation rate in the experimental group was 87.5% and the tumor formation time was (32.5 ± 4.1) d, the difference was statistically significant (P <0.05). The immune function of tumor-bearing mice did not change significantly. The positive rate of CD19 positive cells in tumor-bearing mice was (90.01 ± 3.51)% in experimental group and (87.79 ± 5.62)% in control group with no significant difference (P <0.05). The number of NK cells (40.7 ± 2.2 and 60.3 ± 1.9) in the experimental group and the control group on the 30th day were significantly higher than that of the control group (15.1 ± 0.6) (P <0.05) Group (P <0.05). Conclusion Pristane combined with A20 cell line successfully constructed an animal model of disseminated B-cell lymphoma with certain immune function. The tumorigenesis rate was high, tumorigenesis time was short, and the immune function was retained.
